芥菜是我国重要的鲜食和精深加工蔬菜,其用途包括鲜食、加工、调味、榨油等。其中,我国加工芥菜产业规模庞大,具有重要的经济价值。芜菁花叶病毒(TuMV)是芥菜生产中最重要的病害,导致植株矮化、褪绿,产量与商品品质显著下降。本研究对205份芥菜种质资源进行苗期人工摩擦接种和ELISA检测,鉴定其TuMV抗性。结果表明,205份芥菜种质在抗性上存在显著差异,病情指数分布于0至100之间,共筛选出免疫材料3份、高抗材料16份、抗病材料4份。以发病率和病情指数为变量,采用平方欧氏距离计算材料间遗传距离,利用组间连接法进行系统聚类分析将材料划分5个类别,与抗性分级结果高度吻合。对其中83份代表性芥菜种质进一步开展ELISA检测,发现表型与病毒含量基本一致。抗病种质主要来源于福建、四川、广西等省区,变种类型以笋子芥、大头芥、宽柄芥为主。选出的优异抗性资源可为芥菜抗TuMV新品种培育提供材料基础。
Brassica juncea is an important specialty vegetable and processing material in China, used for fresh consumption, processing, seasoning, and oil extraction. Among these, the processing-type mustard industry has a large scale and irreplaceable economic value. Turnip mosaic virus (TuMV), as the most damaging viral disease in mustard, severely restricts mustard production, causing stunting, chlorosis, and significant reductions in yield and commercial quality, thus constraining the high-quality and sustainable development of the vegetable industry. In this study, 205 mustard germplasm accessions were inoculated by mechanical rubbing at the seedling stage and subjected to ELISA to evaluate their resistance to TuMV. The results showed significant differences in resistance among the 205 accessions, with disease index ranging from 0 to 100. A total of 3 immune, 16 highly resistant, and 4 resistant accessions were screened out. Using incidence rate and disease index as variables, the genetic distance between accessions was calculated based on squared Euclidean distance. Systematic cluster analysis using the between-groups linkage method classified the materials into five groups, which showed high consistency with the resistance grading results. Further ELISA analysis of 83 representative Brassica juncea accessions revealed that the phenotypes were largely consistent with virus accumulation levels. The resistant germplasm mainly originated from Fujian, Sichuan, and Guangxi provinces, with predominant varieties being Brassica juncea var. crassicaulis Chen et Yang, Brassica juncea var. megarrhiza Tsen et Lee, and Brassica juncea var. Latipa Li. The elite resistant sources identified in this study provide a material basis for TuMV-resistant breeding and new variety development in Brassica juncea.
