为明确桂东南地区火力楠(Michelia macclurei)天然群体的表型变异特征、遗传多样性水平及群体遗传结构,并挖掘与重要表型性状相关联的SSR标记,以来源于桂东南地区的5个天然群体共120份火力楠优树资源为试验材料,测定13个表型性状,并利用10对SSR引物进行基因分型,结合聚类分析、群体结构分析和混合线性模型(MLM)开展遗传多样性与关联分析。结果表明,5个火力楠群体在叶片形态和生理生化指标上存在显著差异,其中北流(BL)群体表现出较大的叶片和较高的生物量特征,苍梧(CW)群体的平均丙二醛含量和游离脯氨酸含量显著较高,玉林(YL)与六万(LW)群体表型距离最为相近。10对SSR引物共检测到77个等位基因,平均等位基因数为7.70,平均有效等位基因数为3.72,平均观测杂合度和期望杂合度分别为0.53和0.66,平均多态信息含量为0.62。5个群体中,YL群体遗传多样性最高,高海拔的LW群体最低。群体间平均遗传分化系数为0.11,平均基因流2.89。遗传结构分析表明BL和YL群体保留了相对独立的遗传组分,其余群体表现出较明显的遗传混合。关联分析共检测到6个SSR位点与表型性状显著关联,解释率在6.87%~18.90%之间,其中M23位点与多个性状显著相关。本研究揭示了桂东南火力楠的遗传变异,筛选得到的关联位点可为火力楠种质资源保护、优异种质资源筛选和分子辅助育种提供理论依据。
To clarify the phenotypic variation, genetic diversity, and population genetic structure of natural populations of Michelia macclurei in southeastern Guangxi, and to identify SSR markers associated with important phenotypic traits, this study used 120 superior tree resources collected from five natural populations in southeastern Guangxi as experimental materials. Thirteen phenotypic traits were measured, and genotyping was performed using 10 SSR primer pairs. Cluster analysis, population structure analysis, and a mixed linear model (MLM) were employed to evaluate genetic diversity and marker-trait associations. The results showed significant differences among the five populations in leaf morphological and physiological and biochemical traits. Among them, the BL population exhibited larger leaves and higher biomass, whereas the CW population showed significantly higher average malondialdehyde and free proline contents. The YL and LW populations showed the smallest phenotypic distance. A total of 77 alleles were detected using the 10 SSR primer pairs, with an average number of alleles of 7.70 and an average effective number of alleles of 3.72. The mean observed heterozygosity and expected heterozygosity were 0.53 and 0.66, respectively, and the mean polymorphism information content was 0.62. Among the five populations, the YL population showed the highest genetic diversity, whereas the high-altitude LW population showed the lowest. The average genetic differentiation coefficient among populations was 0.11, and average gene flow was 2.89. Genetic structure analysis indicated that the BL and YL populations retained relatively independent genetic components, whereas the remaining populations showed obvious genetic admixture. Association analysis detected six SSR loci significantly associated with phenotypic traits, with phenotypic variation explained ranging from 6.87% to 18.90%. Among them, locus M23 was significantly associated with multiple traits. This study revealed the genetic variation of M. macclurei in southeastern Guangxi, and the associated loci identified here can provide a theoretical basis for germplasm resources conservation, elite germplasm resources screening, and marker-assisted breeding of M. macclurei.
