    摘要:

    糜子抗旱耐瘠薄,是盐碱地先锋作物。本研究在水培条件下,采用160 mmol/L NaCl溶液对523份国内外糜子种质资源进行苗期耐盐性综合评价。结果表明,盐胁迫下变异明显(耐盐系数小于下四分位数0.65的)表型指标为植株含水率、茎秆含水率、叶面积、根含水率、直径0.2 mm以下根系总长度、直径0.5 mm以上根系总长度。植株干重耐盐系数和植株含水率的耐盐系数与总根长、直径0.2 mm以下根系总长度、直径0.2~0.5 mm根系总长度的耐盐系数均呈极显著正相关,茎秆干重耐盐系数与根系干重、根含水率、总根长的耐盐系数均呈极显著负相关。主成分分析提取的7个主成分累计贡献率达85.198%,其中第1和第3主成分分别由根系直径相关性状耐盐系数及根表面积耐盐系数组成。523份参试糜子种质资源耐盐性综合评价值(D)在0.291~0.654区间内呈正态分布,平均D值较高的种质来源于宁夏(0.493)、青海(0.483)和新疆(0.477),变异系数较大的种质资源来源于山西(16.08%)、甘肃(17.34%)和内蒙古(15.97%)。结合隶属函数将523份糜子种质资源耐盐性划分为5个等级,其中高度耐盐种质资源59份,D值范围0.532~0.654,占比11.28%;中等耐盐种质资源40份,D值范围0.510~0.531,占比7.65%;耐盐种质资源69份,D值范围0.481~0.509,占比13.19%;中等盐敏感种质资源132份,D值范围0.439~0.480,占比25.24%;高度盐敏感种质资源223份,D值范围0.291~0.438,占比42.64%。通过逐步回归分析建立精度为93.55%的耐盐评估模型,确定总根长、植株干重、侧根角度30~60°比例、茎秆含水率、直径0.2 mm以下根系总长度和0.5 mm以上根系总长度的耐盐系数是糜子苗期耐盐性评价关键指标。

    Abstract:

    Broomcorn millet is recognized as a pioneer crop for saline-alkali lands due to its resilience in drought and barrenness. This study conducted a comprehensive assessment of salt tolerance in 523 domestic and international broomcorn millet (Panicum miliaceum L.) germplasm accessions at the seedling stage using a hydroponic system with 160 mmol/L NaCl. Under salt stress conditions, several morphological traits exhibited significant variation (applying salt tolerance coefficient below the lower quartile value of 0.65). These traits included plant moisture content, stem moisture content, leaf surface area, root moisture content, total root length with a root diameter less than 0.2 mm and total root length with a root diameter above 0.5 mm. Correlation analysis further revealed that the salt tolerance coefficient of plant dry weight and plant moisture content are significantly positively correlated with the total root length, total root length with a diameter less 0.2 mm, and total root length with a diameter between 0.2~0.5 mm. In contrast, the salt tolerance coefficient of stem dry weight was significantly negatively correlated with the root dry weight, root moisture content and total root length. Principal component analysis extracted seven principal components, collectively accounted for 85.198% of the total variance. The first and third principal components were predominantly composed of salt tolerance coefficient of root diameter and root surface area, respectively. The comprehensive evaluation values of salt tolerance (D-value) followed a normal distribution, ranging from 0.291 to 0.654. The germplasm resources from Ningxia (0.493), Qinghai (0.483) and Xinjiang (0.477) generally exhibited higher D-value than those from other regions, while germplasm resources from Shanxi (16.08%), Gansu (17.34%) and Inner Mongolia (15.97%) showed higher coefficient of variation. These accessions were classified into five grades based on membership function analysis, including 59 highly salt-tolerant germplasm resources (11.28%, D=0.532~0.654), 40 moderately salt-tolerant germplasm resources (7.65%, D=0.510~0.531), 69 salt-tolerant germplasm resources (13.19%, D=0.481~0.509), 132 moderately salt-sensitive germplasm resources (25.24%, D=0.439~0.480) and 223 categorized as highly salt-sensitive germplasm resources (42.64%, D=0.291~0.438). Through stepwise regression analysis, a salt tolerance evaluation model with an accuracy of 93.55% was established, the key indicators for salt tolerance evaluation of broomcorn millet seedlings were determined to be total root length, plant dry weight, ratio of lateral root angle 30~60°, stem moisture content, total root length with a diameter below 0.2 mm and total root length with a diameter above 0.5 mm.
